响应面法优化泥螺糖胺聚糖提取工艺研究

摘  要:
[目的]探讨采用响应面法优化提取泥螺糖胺聚糖的工艺条件。[方法]以液料比、酶解时间和酶添加量为自变量,利用响应面法对泥螺糖胺聚糖的提取工艺进行优化,建立二次回归方程,确定泥螺糖胺聚糖的最佳提取工艺。[结果]3个因素对泥螺糖胺聚糖提取率的影响依次为:酶添加量>酶解时间>液料比。所建立的二次回归模型中失拟项的P值>0.05,复相关系数为0.996 4,说明该模型具有较高的拟合度。经拟合得到泥螺糖胺聚糖提取的二次回归方程为:Y=26.36+0.22A+0.75B+1.23C-0.052AB+0.097AC-0.390BC-0.72A2-0.47B2-4.10C2。通过响应面法优化,确定泥螺糖胺聚糖的最佳提取工艺为:液料比2(V/W),酶解时间7.5 h,酶添加量0.80%,在此条件下泥螺糖胺聚糖提取率为0.265%。[结论]该研究为泥螺的综合开发利用和多功能糖胺聚糖的制备提供科学依据。

摘  要:
[Objective] The study aimed to discuss the technology conditions for the optimization of extracting glycosaminoglycan from Bullacta exarata by the response surface method.[Method] With the liquid-solid ratio,enzymolysis time and enzyme dosage as the independent variables,the extraction technology for glycosaminoglycan(GAG) from B.exarata were optimized by using the response surface method,the quadratic regression equation was established and the optimum extraction technology for GAG from B.exarata was confirmed.[Result] The effects of 3 factors on the extraction rate of GAG from B.exarata were in order of enzyme dosage > enzymolysis time > liquid-solid ratio.In the established quadratic regression model,the P value in lack of fit was >0.05 and the complex correlation coefficient was 0.996 4,showing that the model had a high fitting degree.The quadratic regression equation for the extraction of GAG from B.exarata obtained through fitting was Y=26.36+0.22A+0.75B+1.23C-0.052AB+0.097AC-0.390BC-0.72A2-0.47B2-4.10C2.After the optimization by the response surface method,the optimum extraction technology for GAG from B.exarata was confirmed as follows: the liquid-solid ratio was 2(V/W),the enzymolysis time was 7.5 h and the enzyme dosage was 0.8%.Under the conditions,the extraction rate of GAG from B.exarata was 0.265%.[Conclusion] The study provided the scientific basis for the comprehensive development and utilization of B.exarata and for the preparation of GAG with multi-function.
