超声诊断在艰难梭菌小鼠感染模型CSS分级评价中的应用

摘  要:
艰难梭菌是常见的院内致病菌之一,在世界范围内均有发生,近年来我国感染病例也有增多的趋势。动物感染模型是研究艰难梭菌的重要工具,本试验将超声诊断技术应用于艰难梭菌感染(CDI)小鼠模型的建立,将超声诊断的图像、参数及结果用于小鼠感染的临床症状严重程度评分(CSS)分级系统中,为艰难梭菌小鼠感染模型评价以及临床诊断和治疗提供超声诊断影像学及数据参考。选用SPF级6~8周龄雄性C57BL/6小鼠,用混合抗生素诱导法建立感染模型。在造模后3 d进行CSS打分、超声诊断及解剖学鉴定。结果表明,CSS系统可以对模型组三类指标进行判定,且感染模型组与对照组呈现极显著性差异(P<0.01)。超声诊断对CDI造成的肠管病变的影像结果,与解剖学鉴定结果一致,解剖后的结肠病变胀气部位直径均值在低无回声区域长和宽均值区间,与长均值呈现极显著性差异(P<0.01),与宽均值相近为无显著差异(P>0.05)。本试验为艰难梭菌小鼠感染模型的建立提供了便捷、实时、准确的评价方法。

摘  要:
Clostridium difficile is one of the common hospital pathogens worldwide.In recent years,the number of infection cases has increased rapidly in China.Animal infection model is an important tool for studying Clostridium difficile.This study applies ultrasound diagnostic technology to the establishment of a mouse model of Clostridium difficile infection(CDI).Ultrasound diagnostic images,parameters,and results were used for the clinical symptom severity score(CSS) grading system for mouse infections,to provide ultrasound diagnostic imaging and data references for the establishment and evaluation of CDI mouse models and clinical diagnosis and treatment.In this study,6-8 weeks old male C57 BL/6 mice of SPF grade were used to establish an infection model using a mixed antibiotic induction method.CSS system,ultrasonic diagnosis and identification of anatomy were performed on the 3 rd day after modeling.The results showed that the CSS system could determine three types of indicators of the CDI model group,and the CDI model group and the control group showed a significant difference(P<0.01).The results of ultrasound diagnosis of intestinal lesions caused by CDI were consistent with the results of anatomical identification.Colon lesions flatulence portion diameter mean was in range of the low echo region length and width mean,the long mean was significant(P<0.01),and width average without significant difference(P>0.05).Therefore,ultrasound diagnosis technology can quickly,effectively determine the infection status of CDI models in vitro.The method not only can determine experimental infection model expectations,estimate the value lesions,but also determine the best treatment and collect real-time feedback treatment results.It could provide a convenient,real-time and accurate evaluation method for the establishment of CDI mouse models.
