大果紫檀试管苗玻璃化影响因子的研究

摘  要:
[目的]研究大果紫檀试管苗出现玻璃化的影响因素。[方法]以大果紫檀的无菌种子苗为材料,研究MS培养基附加不同激素组合及改良MS培养基中NH+4离子、Ga2+离子浓度对大果紫檀试管苗玻璃化的影响。[结果]当TDZ浓度大于0.03 mg/L时,大果紫檀玻璃化比率会大幅上升,而KT不会导致大果紫檀试管苗出现玻璃化;适当降低培养基中NH+4离子浓度及提高Ga2+离子浓度,也能降低试管苗玻璃化发生的比例。[结论]激素种类和浓度是影响大果紫檀试管苗玻璃化的主要因素,低浓度的TDZ既能高效诱导大果紫檀试管苗增殖,又能使试管苗的玻璃化比率处于较低的水平。

摘  要:
[Objective] The aim was to study the factors of causing vitrification for Pterocarpus macarocarpus.[Method] With aseptic seed seedling of Pterocarpus macarocarpus as material,the effects of MS medium supplemented with different hormone combinations and the concentrations of NH+4,Ga2+ in the modified MS medium on the vitrification of regenerated shoots from Pterocarpus macarocarpus were researched.[Result] The results showed that the vitrification ratio of regenerated shoots increased greatly when the concentration of TDZ was above 0.03 mg/L as well as the KT has no effect on vitrification.Properly decreasing the concentration of NH+4 and increasing the concentration of Ga2+ in the medium could reduce the percentage of the vitrification ratio of regenerated shoots.[Conclusion] The different kinds of hormones and their different concentrations were the main factors that affected the percentage of vitrified shoots.Low concentration of TDZ induced shoot to proliferate efficiently,as well as reduced the ratio of vitrification.
