月鳢和乌鳢同工酶的比较研究

摘  要:
[目的]了解月鳢和乌鳢的生化遗传特性,为其品种鉴定提供理论依据。[方法]采用聚丙烯酰胺凝胶垂直梯度电泳法对月鳢和乌鳢的眼、肌肉、心脏、脾、脑和肾中的2种同工酶进行分析。[结果]月鳢肾脏、脾脏、脑和心脏的苹果酸脱氢酶(MDH)图谱有很清晰的4条带,且带型相同、活性相当;其眼组织的MDH图谱有5条带,肌肉组织的MDH带型分布表现出明显活性区。乌鳢肾脏、脾脏、脑、肌肉和心脏的MDH图谱有很清晰的4条带;其眼组织的MDH图谱有5条带。月鳢和乌鳢不同组织中酶的图谱数量一样,但类型不同。在乌鳢6种组织中,眼睛的MDH表达量最大,而月鳢各组织的MDH表达量差异不大。乳酸脱氢酶(LDH)在乌鳢中的表达量高于月鳢。[结论]硬骨鱼类同工酶系统具有明显的组织特异性和物种特异性,其变化可更直接、正确地反映生物体的差异。

摘  要:
[Objective] The purpose was to learn about the biochemical heredity characteristics of Channa asiatica and Channa argus so as to provide theoretical basis for their variety identification.[Method] The 2 isozymes in the eye,muscle,heart,spleen,brain and kidney of C.asiatica and C.argus were analyzed by vertical gradient electrophoresis with polyacrylamide gel.[Result] The maps of MDH from the kidney,spleen,brain and heart of C.asiatica had 4 very clear strips with same strip type and equal activity;that of MDH from its eye tissue had 5 strips and the strip type distribution of MDH from muscle tissue showed obvious active zone.The maps of MDH from the kidney,spleen,brain,muscle and heart of C.argus had 4 very clear strips and that of MDH from its eye tissue had 5 strips.The quantities of maps of the enzyme from different tissues of C.asiatica and C.argus were same,but their types were different.Among the 6 tissues of C.argus,the expression quantity of MDH from its eye was highest,but the expression quantities of MDH from different tissues of C.asiatica had little difference.The expression quantity of LDH in C.argus was higher than that in C.asiatica.[Conclusion] The isozyme system of osteichthyes had obvious tissue and species specificities and its variations could more directly and correctly reflect the difference of organisms.
