AtUGAE4基因反义表达载体构建及对烟草(N.tabacum L.)的转化

摘  要:
本研究从拟南芥(Arabidopsis thaliana)gDNA中克隆了AtUGAE4基因的家族保守序列片段,反向连接在AtHSP70热激启动子和NOS终止子之间,构成含有该基因反义表达盒的表达载体pV-70brUGAE4,并经根癌农杆菌(Agrobacterium tumefaciens)介导,成功导入烟草基因组。这为探讨抑制UGAE家族基因表达对植物果胶生物合成和对离体培养细胞的细胞粘连性的影响奠定了基础。

摘  要:
A conserved sequence of UGAE gene family was cloned from the genomic DNA of Arabidopsis thaliana,and inserted between AtHSP70b heat shock promoter and NOS terminator to form a recombinant plant expression vector pV-70brUGAE4 including an AtHSP70b-reAtUGAE-Tnos expression cassette,which was then successfully transformed into genome of tobacco(N. tabacum L.)mediated by Agrobacterium tumefaciens. The results will provide a possibility to find out how the inhibition on expression of UGAE gene family influences both the biosynthesis of pectin and the intercellular adhesion in vitro of plant cell.
