摘 要：
本文报道了溶菌酶的浓度,酶解温度、时间,生长培养基成分和上再生培养基成分等因素对农抗5102产生菌两个营养缺陷型菌株的原生质体形成和再生的影响。实验结果表明:溶菌酶用量为4mg/ml时,合适的酶解温度和时间是32℃,1.5小时;生长在PM_1培养基和有机A培养基上的菌丝细胞壁对溶菌酶最敏感;原生质体不仅可以在高渗的RM_2培养基上形成菌落,而且也可以在非高渗的PM_1培养基上形成菌落;补充10%蔗糖的PM_1培养基上生长的菌丝可以增加对溶菌酶的敏感性和原生质体的再生频率;当原生质体在4℃中贮存24小时和48小时后,再生频率可分别下降达76%和和94%。

摘 要：
The effects of factors such as enzymc concentration, lytic temperature,lytic time and components of growth medium and regeneration medium 0n theformation and regeneration of protoplasts in two Streptomyces auxtrophs ofantibiotic producing strain 5102 were studied in this paper. The results showed: 1.Optimal lytic temperature and time are 32℃ and1.5 hours at 4 mg/ml enzyme. 2.The cell wall of mycelia grown in the PM:medium or the organic A medium was most sensitive to lysozyme. 3. The pro-toplasts were able to from colonies not only on hypertonic RM_2,but also onnon-hypertonic PM_1 media. 4. Mycelia grown on the PM_1 media supplementedwith 10% sucrose increased the susceptibility to lysozyme and the regenerationratio of protoplasts. 5. After storage of protoplasts at 4℃ for 24 hours and48 hours, the regeneration ratios were reduced more than 76% and 94%, respe-ctively.
