摘 要:
从枯草芽孢杆菌染色体中扩增出受冷诱导的Δ5Des去饱和酶,构建出可以与集胞藻PCC6803染色体发生同源双交换的质粒,将Km抗性基因::PpsbA1::Δ5脂肪酸去饱和酶基因和对照基因分别整合入PCC6803的染色体上,获得转化子后,对不饱和脂肪酸进行研究。结果发现,30℃条件下脂肪酸成分的含量发生了较大变化,20℃时有新成分的出现,且长链脂肪酸的含量有所上升。

摘 要:
Through amplifying the cold-induced Δ5 desaterase from Bacillus subtilis chromosome,the plasmid that could make the homologous double-exchange with the chromosome of Synechocystis sp.PCC6803 was constructed.Km resistant gene(PpsbA1::Δ5 desaterase gene) and control gene were integrated into the chromosome of Synechocystis sp.PCC6803.After obtaining the transformant,the unsaturated fatty acids were studied.The results showed that the content of fatty acids had greater changes at 30 ℃.At 20 ℃,new components was generated and the content of PUFA rose.
