野杏花粉离体培养条件筛选及花粉生活力研究

摘  要:
以野杏优良家系和无性系为研究对象,采用花粉离体培养的方法,对野杏花粉离体培养条件及其生活力进行研究,以期为野杏杂交育种中父本植株的选择提供参考依据。结果表明:培养基组分中蔗糖浓度、培养时间是影响花粉生活力的主要因素。最适宜野杏花粉离体培养的培养基组分浓度为10%蔗糖,0.005%硼酸,培养6.0h。根据野杏花粉管生长速率,可将其划分为4个生长阶段,其中0.3~1.0h为花粉萌发阶段,1.0~2.5h为快速生长阶段,2.5~6.0h为缓慢生长阶段,>6.0h为生长停滞阶段。野杏各家系及无性系均为正常可育,其中家系209号花粉生活力最强,其花粉萌发率及花粉管长度分别为81.33%和1 136.14μm;无性系J118号萌发率最高,花粉管长度最长,分别为63.35%和1 043.78μm。野杏家系花粉萌发率和花粉管长度间无相关性,而无性系呈显著正相关。野杏家系花粉萌发率极显著优于无性系,且有3个家系的花粉生活力均极显著优于西伯利亚杏家系。

摘  要:
Excellent families and clones of Armeniaca vulgaris var.ansu were used as the study objects.The method of pollen culture in vitro was used to study the pollen culture in vitro conditions and pollen vitality of Armeniaca vulgaris var.ansu.In order to provide theoretical basis for selecting the male parent plants in cross breeding of Armeniaca vulgaris var.ansu.The results showed that sucrose concentration and incubation time in the medium components were important factors affecting the pollen viability.The optimum medium of Armeniaca vulgaris var.ansupollen culture in vitro was10% sucrose,0.005% boric acid and cultured for 6.0 hours.It could be divided into four growth periods according to the pollen tubes growth rate,the pollen germination period was 0.3-1.0 hour,the rapid growth period was 1.0-2.5 hours,the slowing growth period was 2.5-6.0 hours and the lag growth period was >6.0 hours.All of Armeniaca vulgaris var.ansufamilies and clones for the normal pollen fertility.The pollen viability of No.209 family was the strongest.Its pollen germination rate and the length of pollen tube were respectively 81.33%,1 136.14μm.The pollen germination rate of No.J118 clone was the highest and with the percent of 63.35.The length of its pollen tube was the longest and with the average length of 1 043.78 μm.There was no correlation between pollen germination rate and pollen tube length of Armeniaca vulgaris var.ansufamilies.The clones were significantly positively correlated.The pollen germination rate of Armeniaca vulgaris var.ansu families were significantly better than clones.The pollen vitality of three families were significantly better than Armeniaca sibiricafamily.
