猪流行性腹泻病毒中国地方流行株LJB/03M基因的克隆与序列分析

摘  要：
从黑龙江某猪场疑似猪流行性腹泻猪的粪便中提取总RNA,采用RT-RCR方法扩增,首次获得中国地方流行株PEDVM全基因序列,将其克隆到pMD18-T载体中进行测序,克隆的M基因核苷酸序列由681个核苷酸组成,含有一个完整的开放阅读框架,编码226个氨基酸。与国外已发表的其他毒株进行基因进化分析,GenBank中的6株PEDV形成的基因进化树具有3个分支。其中分离株LJB/03形成一个独立的分支,说明LJB/03的M基因序列与其他毒株相比,发生了一定的变异。

摘  要：
A porcine epidemic diarrhea virus (PEDV) LJB/03 was isolated from the faece of piglets infected with PEDV on a pig farm, Heilongjiang province, China. The M gene of LJB/03 was amplified from the RNA extracted directly from the faece samples by RT-PCR and cloned into pMD18-T vector. The complete genomic nucleotide sequence was determined by sequencing, and it encompasses an open reading frame of 681 nucleotides, encoding 226 amino acides. Genetic analysis of the M gene, including genetic distance measurement, phylogenetic tree analysis, and residue substitution analysis, showed that the PED viruses fell into three groups, and the LJB/03 itself was as an independent group. These data revealed that the M gene nucleotide sequence of LJB/03 has some mutations compared with the other PED viruses.
