利用RAPD技术分析绵羊品种间遗传关系

摘  要:
利用随机扩增多态性DNA(RAPD)技术研究分析了三种绵羊杂交品种及一种纯种绵羊的基因组DNA多态性。用 10个随机引物对各品种实验羊的基因组DNA进行扩增 ,平均每个品种观察到约 94个标记。扩增结果以RAPDistanceProgram软件分析表明 :中国美利奴羊A品系与中国美利奴羊B品系的随机扩增多态DNA共享度最高 ,它们构成的姊妹群依次与哈萨克羊、中国美利奴羊多胎品系成为对应的姊妹品种。

摘  要:
s: Genomic DNA Polymorphisms in three hybrid sheep and one wild species were detected by using random amplified polymorphic DNA (RAPD) .of 10 decamer nucleotide primers, 9 showed polymorphic banding patterns. On average, about 94 RAPD markers were observed by agarose electrophoresis. The statistical analysis and the clustering tree of the RAPD results done by RAPD istance Program indicated: Chinese Merino strain A and Chinese Merino strain B have the highest ratio of random amplified polymorphic DNA fragments shared, suggesting relationship between them is most closed. In the constructed tree, Chinese Merino strain A and Chinese Merino strain B is the sister group for each other, and then they constitutes a secondary sister group with Kazak(H) Chinese Merino strain D is the last counterpart to the sister group of A and B and H.
