龙牙百合珠芽组织培养研究

摘  要：
[目的]为百合快速繁殖提供参考。[方法]以龙牙百合珠芽为外植体进行组织培养,分别研究外植体消毒条件(75%酒精浸泡20或30 s后用0.1%HgCl2处理8、10、12 min)、珠芽处理方式(整个珠芽、单瓣珠芽、中心几个叶原基、单瓣珠芽上部、单瓣珠芽基部)、激素种类和浓度(100、200、300 mg/LGA、IBA、NAA)对不定芽诱导效果的影响。[结果]75%酒精浸泡20 s后用0.1%HgCl2处理12 min消毒效果最好,外植体污染率较低,成活率较高;以整个珠芽为外植体时,成活率最高,但诱导的不定芽数量较少,单瓣珠芽基部诱导的不定芽数量最多;300 mg/LIBA浸泡珠芽10 h最有利于不定芽诱导,GA对不定芽诱导无明显作用。[结论]该研究确定了龙牙百合珠芽组织培养的最佳条件。

摘  要：
[Objective] The research aimed to provide reference for rapid propagation of lily.[Method] The Lilium brownii var.Viridulum bulblet was used as explant for tissue culture to research the influence of sterilization conditions of explant(treatment with 0.1% corrosive sublimate for 8,10 and 12 min after soaking in 75% alcohol for 20 or 30 s),treatment methods of bulblet(whole bulblet,single-lobe bulblet,several central leaf primordium,upper and base part of single-lobe bulblet),hormone species and concn.(100,200,300 mg/L GA,IBA and NAA) on the induction effect of adventitious bud.[Result] The sterilization effect of treatment with 0.1% corrosive sublimate for 12 min after soaking in 75% alcohol for 20 s was best,the explants reached lower pollution rate and higher survival rate.The survival rate of culture with whole bulblet as explant was highest,but the induced adventitious buds were less.The adventitious buds induced from base part of single-lobe bulblet were most.Soaking in 300 mg/L IBA for 10 h was most favorable for adventitious bud induction from bulblet and GA had no obvious action on adventitious bud induction.[Conclusion] The optimum condition for tissue culture of Viridulum bulblet was confirmed in this research.
