摘 要:
为了探究脂多糖(LPS)刺激仔猪发生炎症时炎性因子肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)、lncRNA-44651和lncRNA-45208在仔猪胸腺、淋巴结和空肠的表达差异,试验选取8头杜×长×大健康断奶仔猪并按照是否注射LPS随机分为对照组和LPS组,各组仔猪在饲喂基础饲粮14 d后,分别按体重100μg/kg腹腔注射生理盐水和LPS,4 h后屠宰仔猪并取胸腺、淋巴结和空肠样品置液氮中保存,采用基因表达测定的方法研究仔猪胸腺、淋巴结和空肠样品相关基因的表达情况。结果表明:LPS刺激4 h后,LPS组仔猪胸腺、淋巴结和空肠中炎性因子TNF-α的mRNA相对表达量均无显著变化(P>0.05),IL-1β的mRNA相对表达量均极显著上调(P<0.01);LPS组仔猪胸腺中lncRNA-44651的mRNA相对表达量极显著上调(P<0.01),lncRNA-45208的mRNA相对表达量显著上调(P<0.05);LPS组仔猪淋巴结中lncRNA-44651和lncRNA-45208的mRNA相对表达量均极显著上调(P<0.01);LPS组仔猪空肠中lncRNA-44651的mRNA相对表达量极显著上调(P<0.01),lncRNA-45208的mRNA相对表达量显著下调(P<0.05)。说明lncRNA-44651和lncRNA-45208可能参与了机体的炎症反应,可为后续仔猪机体功能研究提供候选lncRNA。

摘 要:
To investigate the expression differences of inflammatory factors TNF-α, IL-1β, lncRNA-44651 and lncRNA-45208 in the thymus, lymph nodes and jejunum of piglets when lipopolysaccharide(LPS) stimulated inflammation in piglets, eight healthy weanling Duroc × Landrac × Yorkshire pigletss were randomly divided into control group and LPS group according to whether they were injected with LPS or not. The piglets were fed with basic diet for 14 days, then injected with normal saline or LPS according to their body weight of 100 μg/kg intraperitoneally. Four hours later, the piglets were slaughtered and the thymus, lymph nodes and jejunum samples were collected and stored in liquid nitrogen. The expression of related genes in thymus, lymph nodes and jejunum of piglets was detected by gene expression assay. The results showed that there was no significant change of TNF-α mRNA expression in thymus, lymph node and jejunum(P>0.05) in LPS group and control group after 4 hours of LPS stimulation, and the relative mRNA expression of IL-1β was significantly increased(P<0.01) in LPS group compare with control group, the relative expression of lncRNA-44651 and lncRNA-45208 in thymus was significantly increased(P<0.05) in LPS group. The relative expression of lncRNA-44651 and lncRNA-45208 in lymph nodes of LPS group was significantly increased(P<0.01). The relative expression of lncRNA-44651 and lncRNA-45208 in jejunum of LPS group was significantly up-regulated(P<0.01) and down regulated(P<0.05), respectively. It is suggested that lncRNA-44651 and lncRNA-45208 may be involved in the inflammatory response of the body,which may provide candidate lncRNA for the follow-up study of piglet body function. These results suggestted that lncrna-44651 and lncrna-45208 might be involved in the inflammatory response of the organism, and might provide candidate lncRNA for the subsequent piglet functional studies.
