用RPE冷冻器冷冻小鼠胚胎试验

摘 要:
用RPE冷冻器冷冻42枚桑椹胚和13枚囊胚。冷冻液为1M甘油杜氏磷酸盐缓冲液,胚胎采用0.25ml细管包装。0℃时置入冷冻器。以1℃/分降温,-6.5~-7℃诱发结晶,再以0.5℃/分(0.3~0.8℃/分)降温至-30℃,然后投入液氮。35℃水浴解冻。桑椹胚与囊胚的冻后存活率分别为71.4%(30/42)和53.8%(7/13),差异不显著(P>0.05)。胚胎冻后发育步调很不一致。

摘 要:
42 morulae and 13 blastocysts were frozen in 1 M glycerol/PBS in the RPE freezer. Mouse embryos were loaded into 0.25ml plastic straws that were placed into the freezing apparatus at 0℃. The cooling rate was about 1℃/min from 0℃ to -6.5℃. They were crystallized between -6.5℃ and -7℃ and cooled from -7℃ to -30℃ at 0.5℃/min (0.3-0.8℃/min),then plunged into liquid nitrogen. Thawing was in 35℃ water bath. Post-thaw survival of morulae and blastocysts were respectively, 7l.4%(30/42) and 53.8% (7/13) with no significant difference (P>0.05). The result indicated that post-freezing development of mouse embryos did not keep in step greatly.
