哈萨克马斑点毛色KIT、 MITF基因在皮肤组织的表达

摘  要:
为了研究KIT、MITF基因与哈萨克马斑点毛色的斑点毛形成的相关性,试验选择白斑点骝毛马3匹(包括白斑点1、骝毛)、白斑点栗毛马3匹(包括白斑点2、栗毛)、青毛马3匹采取皮肤组织样品,采用实时荧光定量PCR方法对其皮肤组织中KIT、MITF基因的mRNA表达量进行研究。结果表明:青毛马皮肤组织中KIT基因的表达量最高,与骝毛、栗毛马之间差异显著(P<0.05),栗毛马皮肤组织中KIT基因表达量最低;MITF基因在白斑点1马皮肤组织中表达量最高;骝毛马皮肤组织中MITF基因表达量与青毛马、白斑点1马、白斑点2马、栗毛马皮肤组织中表达量差异显著(P<0.05),骝毛马皮肤组织中表达量最低。说明KIT、MITF基因在不同斑点毛色哈萨克马皮肤组织中mRNA表达量存在差异,白斑点毛马和青毛马皮肤组织中两个基因的表达量均较高。说明哈萨克马斑点毛色的白斑点和青毛的形成与KIT、MITF基因的表达量存在相关性。

摘  要:
In this experiment,real-time fluorescence quantitative PCR method was used to study the mRNA expression of KIT and MITF genesin skin tissues of different colors from Kazakh spotted horse. The test animals included three white ponies bay-hair horses(NO.1 white and bay-hair), three white ponies chestnut-hair horses(NO.2 white-spot and blue-hair), three white ponies blue-hair horses.The results showed that the expression of KIT gene was the highest in blue-hair skin tissue,which had significant difference(P<0.05)with that in bay-or chestnut-hair skin tissues,and it was the lowest in chestnut-hair skin tissue. The expression of MITF gene was the highest in NO.1 white spot skin tissue. Its expression in bay-skin tissue had significant difference(P<0.05)with that in blue-hair,NO.1 white-spot,NO.2 white-spot,or chestnut-hair skin tissues,and was the lowest in bay-skin tissue. The results suggested that significant difference existed on the mRNA expression of KIT or MITF genes of Kazakh spotted horse with different colors. The expression of both genes was high in white-spot or blue-hair skin tissues. The correlation existed between the expression of KIT or MITF genes and white-spot or blue-hair formation from Kazakh spotted horse.
