天然培养基与合成培养基灭菌方式的研究

摘  要:
试验选择5种天然培养基和5种合成培养基。每类培养基分装到20个三角瓶中,天然培养基分为2组(天然培养基Ⅰ组和Ⅱ组),合成培养基分为2组(合成培养基Ⅰ组和Ⅱ组)。天然培养基Ⅰ组、合成培养基Ⅰ组和天然培养基Ⅱ组、合成培养基Ⅱ组,在高压灭菌15min后,分别自然冷却降至压力为0和慢慢放气降至压力为0,然后放置培养箱中培养,并记录污染情况。结果显示,天然培养基适合自然降压灭菌,而合成培养基适合慢慢放气降压灭菌。

摘  要:
Five kinds of natural medium and five kinds of synthetic medium were selected. Each medium was dumped into 20 conicalflasks. Each media had two treatments. In the first treatment, the natural media and synthetic media were sterilized for 15 min under maintaining pressure and naturally cooled until zero pressure. In the second treatment, the natural media and synthetic media were sterilized for 15 min under maintaining pressure and then the pressure was decreased to zero by releasing steam. All sterilized media were incubated in an incubator to observe contamination. The results showed that natural depressurization is suitable for sterilization of natural medium, while releasing steam is suitable for sterilization of synthetic medium.
