一起麻鸡皮肤型鸡痘的诊断及病原分离鉴定

摘  要:
采用PCR检测、动物回归试验、病原分离鉴定、病理组织学观察和间接免疫荧光检测等方法,对一起疑似鸡痘病毒(FPV)感染的临床病例进行了系统的分析。结果表明:PCR扩增结果为阳性;将病料处理上清液经尿囊膜途径接种SPF鸡胚,可见尿囊膜上形成有灰白色突起的痘斑;将分离获得的病毒以背部接种1月龄的SPF雏鸡,感染雏鸡表现精神萎靡,食欲减退,在接种部位出现典型的痘疹,出现与自然感染病例相类似的临床症状;采集攻毒感染SPF雏鸡各组织器官进行病理组织学观察和IFA检测,仅在感染部位皮肤的胞浆内见有特异性的嗜酸性病毒包涵体及绿色荧光信号,而其他组织脏器未见有阳性反应。确诊该起病例为鸡痘病毒感染所致的皮肤型鸡痘。

摘  要:
Eruptions of poxes of different sizes and nodular lesions were observed in the skins of crista galli,eyelid and the smoothness and few feather of the back.In severe case,the thick scabs were observed because of the fusion of pox eruptions.According to the typical clinical symptoms,we diagnosed the disease as skin type fowlpox.In order to make final diagnosis for the disease,we carried on systematic analysis by PCR detection,animal regression experiment,isolation and identification,pathohistology observation and IFA detection.The PCR results showed that the amplification result was positive using specific primers of FPV p4b core protein gene.We inoculated the processing supernanant of pox eruptions on the surface of chorioallantoic membrane of 9-day-old SPF embryonated egg.After 5 days,grayish-white variola were observed on the surface of chorioallantoic membrane.Then,SPF chickens aged 1 month were inoculated by back acupuncture route using the obtained FPV virions.After 6 days,the chickens of experiment group showed dispirited,decreased food appetite,and typical pox eruptions were observed in the sites of innoculation.The tissues of inoculated SPF chickens were collected in order to carry on pathohistology observation and IFA detection.The specific intracytoplasmic eosinophilic inclusion bodies and green fluorescence were detected in the epithelium,respectively.The positive reaction was not observed in other tissues.Based on the laboratory detection results and the typical symptoms,we finally identified that the case was skin type fowlpox caused by fowlpox virus(FPV) infection.
