枣庄石榴种质资源遗传多样性的ISSR分析

摘  要:
以39份枣庄石榴品种为试材,采用ISSR标记方法,研究了石榴品种间的遗传多样性及遗传关系,以期为枣庄石榴种质资源保存、杂交育种奠定基础。结果表明:9条ISSR引物对39个品种进行扩增,共检测到168个DNA位点,平均每条引物扩增出18.67个DNA位点,其中多态性位点156个,多态性位点比率(PPB)为92.86%。供试样品平均观察等位基因数(Na)、有效等位基因数(Ne)、Nei′s基因多样性指数(He)及Shannon信息指数(I)分别为1.928 6、1.690 9、0.384 7、0.557 3;品种的遗传相似系数(GS)在0.410 7~0.958 3,采用UPGMA构建遗传关系聚类图,以遗传相似性系数(GS)0.656为阈值,供试的39份石榴品种可分为5类,根据ISSR标记的品种分类结果与根据形态性状分类结果不一致。

摘  要:
The 39 pomegranate cultivars from Zaozhuang were used as experimental materials in this study.The genetic relationships and genetic diversity of 39 pomegranate cultivars were analysed by ISSR markers in order to provided the foundation on the storage and breeding of the pomegranate resources at Zaozhuang.The results showed that a total of 168 loc were amplified from 9 ISSR primers with an average of 18.67 bands from a primer,of which 156 were polymorphic loci.The main indexes were,that percentage of polymorphic band(PPB)=92.86%,the number of alleles(Na)=1.928 6,Effective number of alleles(Ne)=1.690 9,Nei′s gene diversity(He)=0.384 7 and Shannon′s information index(I)=0.557 3.The genetic similarities coefficient(GS)between cultivars ranged from 0.410 7 to 0.958 3,39 pomegranate cultivars were divided into 6 groups at GS 0.656 based on UPGMA method.The results of ISSR cluster did not accord with the classification results based on some morphological variations.
