基于BSA法的甜瓜果肉酸味相关性状主效QTL分析

摘  要:
以果肉有酸味甜瓜品系MR-1为母本,果肉无酸味甜瓜品系M1-15为父本,配制杂交组合获得F2代,并自交获得F2:3代群体.通过分析甜瓜果肉性状相关性,发现果肉酸味与果肉pH呈显著负相关.利用群体分离分析法(BSA)筛选发现调控甜瓜果肉酸味基因位于第5号和8号染色体.基于亲本材料作高通量测序,在候选基因组区间开发40对SNP-CAPS引物,经筛选26对在亲本间具有多态性,多态率为65%.所构建遗传连锁图包含15对引物,2个连锁群,覆盖基因组总长度为140.45 cM,标记间平均距离为9.36 cM.检测到两个与果肉酸味相关的QTL和一个与果肉pH相关的QTL.

摘  要:
F_(2:3) generation was constructed by self-pollinating the resultant F_2 generation from a cross between the melon line MR-1 which had sour flesh as the female parent and M1-15 which had non-sour flesh as the male parent. According to the correlation analysis of melon flesh traits, the fruit sourness trait was negatively correlated with pH. Using the bulked-segregant analysis(BSA) technique to determine the fruit sourness gene on chromosome 5 and chromosome 8. Based on the high-throughput resequencing date of parental materials, 40 pairs of SNP-CAPS primers were designed in candidate genome intervals and 15 pairs polymorphic primers were selected, with a polymorphic rate of 65%. The linkage map constructed with these markers covered two linkage groups, with a total genetic distance of 140.45 cM and mean distance between markers of 9.36 cM. Total three QTLs were detected for flesh sourness and p H with different genetic loci on various chromosomal locations.
