黑羽番鸭PRL、PRLR基因序列分析及表达规律研究

摘  要:
催乳素(Prolactin,PRL)由动物垂体前叶分泌,与其受体(Prolactin receptor,PRLR)相结合才能作用于靶细胞。为了解PRL与PRLR基因在黑羽番鸭不同繁殖阶段各组织内的时空表达规律,利用RACE扩增方法获得了PRL与PRLR基因的CDS区序列,同时应用荧光定量与免疫组化相结合的方法,对产蛋前期、产蛋期、就巢期的黑羽番鸭垂体、卵巢、子宫中PRL与PRLR基因的表达量进行了比较分析。结果显示:PRL与PRLR基因序列在多种禽类中高度同源;PRL基因在垂体、卵巢、子宫中m RNA的表达具有明显的空间特异性,PRL基因在垂体中表达量最高,极显著高于卵巢和子宫中的表达量(P<0.01),这种趋势贯穿产蛋前期、产蛋期和就巢期。PRLR基因在不同时期各组织内m RNA的表达具有明显的时空特异性。PRLR基因的表达量在垂体、卵巢中的表达量普遍较低,子宫中的表达量最高。PRLR基因在不同时期垂体和卵巢中的表达量没有明显的差异(P>0.05),而子宫中的表达量在就巢期显著高于产蛋前期与产蛋期(P<0.05)。

Prolactin(PRL) is secreted by the anterior pituitary of animals, which must be combined with its receptor(Prolactin receptor, PRLR) to act on target cells. In order to understand the expression patterns of PRL and PRLR in the tissues at different reproduction stage of black Muscovy duck, the CDS region sequences of PRL and PRLR were obtained by RACE amplification. The expression levels of PRL and PRLR in pituitary, ovary and uterus of black Muscovy duck at the early stage of laying cycle, laying period and broody period were analyzed by using the combination method of fluorescence quantitation and immunohistochemistry. The results showed that the sequences of PRL and PRLR were highly homologous in a variety of poultry; The m RNA expression of PRL gene in pituitary, ovary and uterus had obviously spatial specificity. The expression of PRL gene in pituitary was the highest, which was significantly higher than that in ovary and uterus(P<0.01). This trend continued for the early stage of laying cycle, laying period and broody period. The m RNA expression of PRL gene in various tissues at different reproductive period had obvious time-space specificity. The expression level of PRLR gene in pituitary and ovary was generally low, and which in uterus was the highest. There was no significant difference in the expression of PRLR gene between pituitary and ovary at different periods(P>0.05), but the expression level of PRLR gene in uterus reached the highest in broody period, which was significantly higher than that at the early stage of laying cycle(P<0.05).
