摘  要:
人工合成一双对应于IBV-Beaudette株N基因ORF两侧序列的引物,应用RT-PCR方法,获得肾型传染性支气管炎病毒Australia-T株核衣壳蛋白基因,长度1.23kb,利用引物设计中的EcoR I位点将其克隆至载体pSK(+)中,对该基因进行限制性酶切分析,结果与已报道的相一致。通过N基因的C端部分序列分析及与Beaudette株、M41标准株和日本分离株相比较,结果表明该部分的序列有一定的差异,其同源率分别为86.8%、86.8%和87.3%,推测的氨基酸同源率分别为87.2%、87.2%和88.7%。

摘  要:
Based on the reported sequence of the nucleocapsid protein gene, IBV Beaudette,two specific primers were synthesized. A 1.23 kb DNA fragment was amplicatedby RT-PCR using the viral RNA of IBV (strain Australia-T) as the template,and furthercloned into plasmid PSK (+). Restriction mapping revealed that the result was consistentwith the reported data. Comparison of sequence of N gene of Australia-T strain with otherIBV strains showed that the homologies were 86. 8%, 86. 8% and 87. 3% in nucleotide and87. 2 %, 87. 2%and 88. 7% in amino acid,respectively.
