版纳微型猪近交系LRWD1基因克隆及功能生物信息学分析

摘 要:
LRWD1基因在精子形成过程中起重要作用。从Gen Bank下载猪及近缘物种的LRWD1 mRNA序列作为参考序列,设计特异引物扩增版纳微型猪近交系(BMI)LRWD1基因。对蛋白质序列进行功能生物信息学分析,构建多物种系统进化树。研究获得了BMI LRWD1 1 944 bp的编码区序列(Gen Bank登录号:KU705639,对应的氨基酸登录号:AOC89057),编码647个氨基酸,蛋白质分子量(Mw)为71.40 k D,等电点(p I)为7.52。功能生物信息学分析表明LRWD1蛋白质存在2个保守结构域,无跨膜结构,无信号肽序列;N末端和C末端均疏水;有6类功能活性位点,位于细胞质的概率是70.6%。系统进化分析表明,BMI与狗、大熊猫的亲缘关系最近。研究结果将为进一步研究LRWD1基因在猪精子发生方面的作用及功能奠定基础。

摘 要:
LRWD1 gene plays an important role in the process of spermatogenesis. Using the LRWD1 mRNA sequences of pig and related species from Gen Bank as reference gene sequences,we designed specific primers and amplified Banna mini-pig inbred line( BMI) LRWD1 gene. Protein sequence was used to carry out functional bioinformatics analysis and a phylogenetic tree was constructed. A coding sequence of 1 944 bp( Gen Bank accession No. KU705639,the corresponding amino acid sequence acces-sion number: AOC89057) of BMI LRWD1 was obtained,which encodes a protein of 647 amino acids,with molecular weight 71. 40 k D,and isoelectric point 7. 52. Functional bioinformatics analysis indicated that the LRWD1 protein contained two conserved domains,with no transmembrane region and signal peptide sequences,and its N-terminal and C-terminal were hydrophobic. This protein has six kinds functional active sites and its 70. 6% located in cytoplasmic. Phylogenetic analysis using LRWD1 sequences demonstrated that BMI had the closest relationship with dog and panda. The results of the study will lay a foundation for further study of the gene about its functions in pig sperm formation.
