基于LC-MS/MS技术对CpA感染鸭十二指肠代谢组学分析

摘  要:
为探讨鸭感染A型产气荚膜梭菌(Clostridium perfringens alpha toxin,CpA)后十二指肠代谢物变化,寻找潜在生物标志物及与致病机制相关的代谢通路,将12只37日龄鸭随机分为对照组和CpA感染组,每组6只,运用液相色谱-串联质谱(LC-MS/MS)技术对对照组和CpA感染90 h的鸭十二指肠进行非靶向代谢组学分析.结果表明:在偏最小二乘判别分析(PLS-DA)模型中,对照组与CpA感染组的得分图呈现明显差异,CpA感染组中部分代谢物发生显著性改变.CpA感染90 h组鉴定出花生四烯酸、3-羟基丁酸、2,3-二羟基苯甲酸、二磷酸腺苷等29种差异代谢物,主要涉及不饱和脂肪酸的生物合成、β-丙氨酸代谢、花生四烯酸代谢、硫代谢、丙酮酸代谢、嘌呤代谢等25条信号通路.CpA感染组与对照组鸭十二指肠之间存在脂类代谢、氨基酸代谢及碳代谢等的差异.研究结果可为CpA感染鸭引起十二指肠损伤机制的研究提供依据.

摘  要:
To investigate the changes of duodenal metabolites of ducks infected with Clostridium perfringens alpha toxin(CpA),and to search for potential biomarkers and metabolic pathways related to the pathogenesis,twelve 37-day-old ducks were randomly divided into control group and CpA infected group with 6 ducks in each group.Liquid chromatoc-tandem mass spectrometry(LC-MS/MS)was used to analyze the non-targeted metabolomics of duodenum in normal group and 90 h CpA infected ducks.The results showed that in partial least squares discriminant analysis(PLS-DA)model,the score maps of control group and CpA infection group showed significant differences,and some metabolites in the CpA infection group had significant changes.After 90 h of CpA infection,29 different metabolites such as arachidonic acid,3-hydroxybutyric acid,2,3-dihydroxybenzoic acid and adenosine diphosphate were identified,and 25 signaling pathways were mainly involved in unsaturated fatty acid biosynthesis,β-alanine metabolism,arachidonic acid metabolism,sulfur metabolism,pyruvate metabolism and purine metabolism.There were differences in lipid metabolism,amino acid metabolism and carbon metabolism of duodenum between CpA infected group and control group.The results of this study could provide a new basis for the study of duodenal injury mechanism caused by CpA infection in ducks.
