1株抗小麦病原真菌活性的射干内生真菌的筛选鉴定及其聚酮合酶基因分析

摘  要:
【目的】挖掘有用的射干内生真菌资源,并为其合理利用提供一定的参考依据。【方法】从射干根中分离获得了1株内生真菌,采用形态学和分子生物学手段鉴定其属种,采用菌丝生长速率法测定其发酵液对禾谷镰刀菌和小麦根腐离蠕孢的抑菌作用,并分析该菌株的聚酮合酶基因序列和生物学特性。【结果】结合形态学和ITS序列同源性分析将菌株鉴定为层出镰刀菌(Fusarium proliferatum)。该菌株的发酵液对禾谷镰刀菌和小麦根腐离蠕孢的菌丝生长有较好的抑制作用。SG5菌株的PKS氨基酸序列与Gen Bank所报道的层出镰刀菌的PKS序列相似性为99.7%。菌株最适生长的固体培养基为察氏培养基,最适碳源为蔗糖,最适氮源为硝酸钾,最适pH为7;在NaCl质量分数≤1%的培养基上正常生长。【结论】该拮抗内生真菌是潜在的小麦病原真菌生防菌资源,具有一定的开发与应用价值。

摘  要:
[Purpose]To excavate useful endophytic fungi resources of Belamcanda chinensis and provide some reference for their rational utilization.[Methods]An endophyte isolate Fusarium proliferatum SG5, was obtained from the roots of B. chinensis. The strain was identified by using internal transcribed spacer sequencing and morphological features, and the antifungal activities of fermentation broth of SG5 stain against two wheat pathogenic pathogens, including F. graminearum and Bipolaris sorokinianum Sacc.ex Sorok, were evaluated by the method of mycelial growth. The PKS sequence as well as some biological characteristics of the strain were also measured. [Result]The SG5 strain was identified as F. proliferatum based on morphological characteristics and ITS sequence analysis. The fermtenation broth of SG5 strain had antagonistic activities against F. graminearum and B. sorokinianum. The amino acid sequence of PKS showed 99.7% identifies to the known F. proliferatum PKS gene sequence in the GenBank. The study on the biological characteristics exhibited that the proper growth agar of the strain was on Czape's medium, and suitable carbon sources was sucrose while the optimal nitrogen source was potassium nitrate. The proper growth pH was 7. In addition,strain show tolerance to 1% NaCl.[Conclusion]This study indicates the SG5 strain is potential biocontrol resources of wheat pathogenic pathogens, and have certain value of development and application.
