德化黑鸡黑素皮质素受体1(MC1R)基因单核苷酸多态性分析

摘  要:
采用聚合酶链式反应及单链构象多态(PCR-SSCP)技术以及DNA直接测序的方法,对德化黑鸡MC1R基因编码区进行单核苷酸多态性(SNPs)分析。结果表明:引物P2扩增片段具有多态性,并表现为AA、AB和BB等3种基因型;经序列分析发现,BB型存在2个突变位点:分别在1094处(G-A)和1095处(T-C),其中在1095处的突变还导致氨基酸的改变(Cys-Arg)。由基因型分布情况表明,德化黑鸡C系(乌色)和D系(非乌色)均以AA型为主;BB型(含SNP)仅存在于C系中,而在D系中未发现,推测BB型可能是乌色性状特有或与乌色性状相关。

摘  要:
Single strand conformation polymorphism analysis of polymerase chain reaction products(PCR-SSCP) and DNA sequence were used to analyze the single nucleotide polymorphisms(SNPs) in MC1R gene exon of Dehua Black Chicken.The results showed that there were polymorphisms in amplified fragment of primer P2,which were AA,BB and AB genotypes.Two single mutation points in 1 904 bp(G-A)and 1 905 bp(T-C)were detected in the BB genotypes,and the mutation T-1 905-C caused an amino acid substitution(Cys-Arg).Furthermore,distribution of the genotypes indicated that the AA genotype was predominant in the two lines of C(the black characteristic) and D(the non-black characteristic).The BB genotype(including SNP) was only detected in line C,not in line D.It was supposed that the genotype BB might be particular or related to the black characteristic of Dehua Black Chicken.
