摘 要:
收集系统性硬化症(SSc)患者血液样本58例,正常人60例,分离外周血单个核细胞(PBMC),通过Real-time PCR方法检测斑联蛋白(ZYX)基因表达并对不同亚型SSc患者中ZYX基因表达量进行分析;利用Ⅰ型干扰素处理PBMC,通过Real-time PCR方法检测ZYX基因表达的变化,研究ZYX在SSc中的表达及其对炎症的作用.结果表明:与正常对照相比, SSc患者PBMC中ZYX的表达显著升高;ZYX在ATA阴性、ACA阳性、关节不累积、食管不累及和心脏不累及的SSc患者中表达水平高于ATA阳性、ACA阴性和器官累及的患者.Ⅰ型干扰素处理可显著降低PBMC中ZYX基因表达;临床中药复方治疗后SSc患者PBMC中ZYX表达降低.这一研究提示ZYX可能与SSc疾病程度和炎症免疫密切相关.

摘 要:
To investigate the expression of zyxin(ZYX) in systemic sclerosis and its effect on inflammation, blood samples were collected from 58 systemic sclerosis patients and 60 normal controls. Peripheral blood mononuclear cells(PBMC) were isolated, and ZYX gene expression was detected by Real-time PCR and ZYX gene expression was analyzed in different subtypes of SSc patients. PBMC was treated with type I interferon and change of ZYX gene expression in cells was detected by Real-time PCR. Compared with normal controls, the expression of ZYX in PBMC of SSc patients was significantly increased. The expression of ZYX in SSc patients with ATA negative, ACA positive, joint non-accumulation, esophageal non-accumulation and cardiac non-accumulation was higher than that of SSc patients with ATA positive, ACA negative, and organ involvement. Type I interferon treatment significantly reduced the expression of ZYX in PBMC, and the expression of ZYX in PBMC of SSc patients treated with traditional Chinese medicine compound was decreased. ZYX may be closely related to SSc disease severity and inflammatory immunity.
