牦牛KISS-1基因的克隆及其在生殖轴的表达

摘  要:
KISS-1在动物繁殖调控中具有重要作用,旨为探讨KISS-1基因在牦牛季节性繁殖中的调控作用。实验采集5头成年母牦牛和5头黄牛的下丘脑、垂体等组织,利用RT-PCR和q PCR技术研究其KISS-1基因序列及组织表达特性。结果表明,牦牛和黄牛KISS-1基因编码区长度为408 bp,编码135个氨基酸,比对分析发现牦牛和黄牛基因编码区存在7处碱基突变。牦牛KISS-1基因氨基酸序列与黄牛、藏山羊、绵羊、野猪、人和褐家鼠的同源性分别为97%、85%、65%、55%和51.5%。KISS-1基因m RNA在牦牛和黄牛的下丘脑、垂体、卵巢、输卵管及子宫中均有表达。在下丘脑和脑垂体的表达丰度高,但两物种间无显著性差异(P>0.05)。说明KISS-1基因在动物进化中比较保守,对牦牛季节性繁殖的调控作用有待进一步研究。

摘  要:
KISS-1 plays a pivotal role in the regulation of animal reproduction,thus this study is aimed to investigate the role of KISS-1 gene in regulating seasonal reproduction of the yak. Samples of hypothalamus,pituitary etc. were collected from five adult female yaks andfive yellow cattle. KISS-1 gene sequence and its tissue expression characteristics were studied by RT-pcr and q PCR techniques. The resultsshowed that the coding region length of yak and yellow cattle KISS-1 gene was 408 bp,which encoded 135 amino acids,and there were 7 base mutations between these two species. The amino acid sequence identities of yak KISS-1 gene with yellow cattle,Tibetan goats,sheep,wild pig,human,and sewer rat were 97.1%,85%,65%,55%,and 51.5% respectively. Expression of KISS-1 gene was detected inhypothalamus,pituitary,ovary,fallopian tube,and uterus of yak and cattle. The expression levels in hypothalamus and pituitary were thegreatest,but there was no significant difference between the two species(P > 0.05). It is concluded that KISS-1 gene is conservative inanimal evolution,and its regulation roles in seasonal reproduction of the yak need to be further studied.
