副结核分枝杆菌单克隆抗体杂交瘤细胞系的建立和鉴定

摘  要:
本试验通过6次细胞融合,经过3次反复克隆化与长时间培养和两次液氮中冻存、复苏,获得了10株稳定、高效地分泌抗副结核分枝杆菌单克隆抗体的杂交瘤细胞系。这些杂交瘤细胞的染色体数量均较其两种母源细胞有明显地增加,但少于两者之和;经培养产生的含单克隆抗体的培养上清液效价在1/100~1/300之间,腹水效价达万倍以上。这10株单克隆抗体都是分枝杆菌特异性的,但在分枝杆菌之间却有广泛的交叉反应。

摘  要:
The ten hybridomas cell lines that produce monoclonal antibodies against mycobacterium paratuberculosis have stably highly been obtained after six-time cell-fusion, through three clonings, by freezing twice in liquid nitrogen or necovery and cultivation for long time. The number of chromosomes of the hybridomas cells is more than that of cells origined hybridomas, but it is fewer than the sum of chromosomes of spleen cell and myeloma cell. The titir of supernatant of the hybridomas attained is 1/100-1/300 and that of ascitic fluids is abovel/10000. The ten monoclonal antibodies are the specific for mycobaeterium, but they have anextensive cross reaction among mycobacteria.
