棉铃虫β-catenin基因的克隆、表达分析及真核表达载体构建

摘  要:
β-catenin基因是生物进化上高度保守的基因,是Wnt/β-catenin信号通路中的重要组分。本研究通过RACE方法获得了棉铃虫β-catenin基因的全长c DNA序列,命名为Har-β-catenin(Gen Bank登录号为KJ206237)。其开放阅读框长2382 bp,编码793个氨基酸残基。与已报道的其它物种β-catenin蛋白相似,Har-β-catenin蛋白具有多个ARM重复结构域。运用RT-PCR的方法比较滞育和非滞育蛹脑中Har-β-catenin基因的表达情况,结果表明非滞育蛹脑中Har-β-catenin基因的表达水平高于滞育蛹脑。最后,我们构建了包含Har-β-catenin全长编码区的真核表达载体并调查了Har-β-catenin的亚细胞定位情况。这些结论为我们进一步研究Wnt/β-catenin信号通路在棉铃虫滞育中的作用奠定了基础。

摘  要:
β- catenin is a highly conserved gene,it is an important component of Wnt / β- catenin signaling pathway. In this study,we cloned the β- catenin c DNA from the Helicoverpa armigera brain.The sequence was named as Har- β- catenin( Gen Bank accession number: KJ206237). Its open reading frame was 2382 bp in length,encoding a 793- amino- acid protein. Multiple conserved ARM( Armadillo /β- catenin- like) repeats,which are a common feature of β- catenin,were found in Har- β- catenin. The mRNA levels of Har- β- catenin were significantly higher in non- diapause-pupal brains than in diapause- destined ones. And finally,we constructed eukaryotic expression vector of Har- β- catenin and studied its subcellular localization. This study lays the foundation for the further functional study of Wnt / β- catenin in H. armigera pupal diapause.
