HRV间接ELISA检测方法的建立及大庆市区腹泻患儿HRV感染率的调查

摘 要:
为了监测近期大庆市儿童人源轮状病毒(HRV)的感染情况,利用已分离鉴定好的一株HRV当地流行株,对该HRV流行株进行浓缩纯化后,以纯化的HRV作为包被抗原,按照设定的间接ELISA程序进行试验,确定并优化各种检测条件,从而建立HRV间接ELISA检测方法;利用建立的方法对2011年7月—2012年3月期间大庆地区1 452例腹泻患儿中的HRV感染情况进行初步调查,并按季节、年龄、区域、性别等方面对HRV感染率的检测结果进行统计学分析。结果显示,建立的HRV间接ELISA检测方法敏感度高、特异性强、重复性好,且检测结果灵敏、准确;利用该方法检测大庆市区1 452例腹泻患儿中HRV感染率平均为39.9%,而且得出寒冷的秋冬季是HRV感染的高发期,婴幼儿为HRV感染的易感时期;此外,卫生条件、经济状况及机体免疫力等因素也与HRV感染有一定的相关性。

摘 要:
In order to monitor human rotavirus(HRV) infection with children in daqing city recent,a local epidemic strains of HRV that Had been separated and identified was used,then differential centrifugation was performed to concentrate and purify the HRV.The purified HRV serving as coating antigen,according to the indirect ELISA procedures set test in order to determin and optimize the optimal conditions of this method.Thus an indirect ELISA method was established to detect HRV.Preliminary epidemiological study was performed on infection with rotavirus in 1 452 children with diarrhea in Daqing between July 2011 and March 2012,using the indirect ELISA diagnostic method established in this trial,and the statistical analysis was performed on HRV infection rates test results according to season,children's age,region,gender etc.The results showed that the indirect ELISA method established in this trial had high sensitivity,high specificity and good repeatability,and the results is sensitive and accurate.1 452 cases of children of the diarrhea was detect using the indirect ELISA diagnostic method established in this trial.The results showed that the infection rate of HRV was 39.9% at average,and indicating that children were susceptible to HRV in the autumn and the winter,and children below 3 years were susceptible to HRV.In addition,HRV infection had a certain correlation with Health conditions,economic status and immunity etc.
