营养组合物对再障大鼠干细胞增殖的影响

摘  要:
目的:观察营养组合物对再生障碍性贫血大鼠干细胞增殖的影响,探讨其治疗再障的可能机制。方法:SD大鼠100只,随机分为正常对照组、再障模型组(AA)及不同剂量的营养组合物组。构建再障模型的同时,第5天开始,分别对大鼠给予高剂量营养组合物2 266.95 mg/(kg·d)、中剂量高剂量营养组合物1 511.3 mg/(kg·d)、低剂量高剂量营养组合物1 057.91 mg/(kg·d),每天灌胃,直至实验结束(60d),采用颈椎脱臼法处死大鼠,采用免疫组化法检测再障大鼠的肝、肾、胃、肠、脑干细胞标志物的表达情况。结果:大鼠肝脏、肾脏及胃的干细胞标志物的含量增加,且随着营养组合物剂量增加而增加;营养组合物处理再障大鼠后,大鼠肠粘膜、神经干细胞的标志物表达量在各组中无显著变化。结论:营养组合物对再障大鼠肝脏、肾脏及胃的干细胞增殖具有一定的促进作用;而对再障大鼠肠粘膜、神经的干细胞增殖无显著影响。营养组合物一定程度上可以促进再障大鼠的干细胞增殖,此可能为其治疗再生障碍性贫血的作用机制之一。

摘  要:
Objective To observe the effect of nutrient mixture on the proliferation of stem cells in aplastic anemia( AA) rats,and explore the possible mechanism of nutrient mixture in treatment of AA. Method Totally 100 SD rats were divided randomly into three groups including normal control,AA model and the different doses of the nutrients mixture groups. AA rat models were built. Five days later,the rats of nutrient mixture groups were fed by nutrients mixture of different doses including 2 266. 95,1 511. 3,1 057. 91 mg/( kg·d),killed by cervical dislocation 60 days later. Then the expression of liver,kidney,stomach,intestine and brain stem cell markers were detected by immunohistochemistry staining. Result After the AA rats treated with the nutrient mixture,the expression of liver,kidney and stomach stem cell markers were increased with the dose of the nutrient mixture. There were no significant changes in the expression of markers of intestinal mucosal stem cells and neural stem cells. Conclusion Nutrient mixture had a certain role in promoting proliferation of liver,kidney and stomach stem cells in AA rats. However,nutrient mixture had no significant effect on the proliferation of intestinal mucosal and neural stem cells in AA rats. Taken together,nutrient mixture promoted stem cells proliferation in AA rats. This might be a mechanism for the treatment of AA.
