细菌裂解蛋白SALy102对伪中间型葡萄球菌的抗菌活性分析

摘  要:
伪中间型葡萄球菌是犬、猫皮肤、耳道和伤口感染的主要病原菌之一,其耐药情况愈发严重且出现多重耐药性状.细菌裂解蛋白是一种新型抗菌物质.为探讨细菌裂解蛋白SALy102对伪中间型葡萄球菌的抗菌效果,本试验从皮肤或耳道感染的患犬中分离鉴定伪中间型葡萄球菌,采用比浊法测定SALy102的抗菌活性,绘制SALy102的时间-杀菌曲线;采用微量肉汤稀释法测定SALy102对伪中间型葡萄球菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC).结果显示,共分离鉴定出12株伪中间型葡萄球菌;SALy102对12株伪中间型葡萄球菌临床分离菌株均具有非常强的抗菌活性;时间-杀菌曲线显示,低浓度(15.63 μg/mL)的SALy102也可以在1 h内快速裂解伪中间型葡萄球菌;SALy102对伪中间型葡萄球菌的MIC为32 pg/mL,MBC为64 μg/mL.结果表明,SALy102对伪中间型葡萄球菌抗菌作用显著,具有临床治疗犬、猫伪中间型葡萄球菌感染的潜力.

摘  要:
Staphylococcus pseudintermedius is one of the major pathogens causing skin,ear,and wound infections in dogs and cats,and its resistance is becoming increasingly serious,with the emergence of multidrug-resistant traits.Bacterial lysin is a novel antimicrobial substance.To investigate the antibacterial effect of bacterial lysin SALy102 against Staphylococcus pseudintermedius,this study isolated and identified Staphylococcus pseudintermedius from infected dogs with skin or ear infections.The antibacterial activity of SALy102 was determined using the turbidity method,and time-kill curve of SALy102 was plotted.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of SALy102 against Staphylococcus pseudintermedius were determined using the microbroth dilution method.The results showed that 12 strains of Staphylococcus pseudintermedius were isolated and identified.SALy102 exhibited very strong antibacterial activity against all 12 clinical isolates of Staphylococcus pseudintermedius.The time-kill curve demonstrated that even at a low concentration(15.63 μg/mL),SALy102 could rapidly lyse Staphylococcus pseudintermedius within 1 hour.The MIC of SALy102 against Staphylococcus pseudintermedius was 32 μg/mL,and the MBC was 64 pg/mL.These results indicate that SALy102 has significant antibacterial effects against Staphylococcus pseudintermedius and holds potential for clinical treatment of Staphylococcus pseudintermedius infections in dogs and cats.
