人工饲养加州海狮MHC-DQB氨基酸序列分析

摘  要:
通过利用克隆与测序技术,分析人工饲养条件下加州海狮的MHC-DQB基因野生型以及其他哺乳动物序列的差异。通过序列比对和进化树分析,发现α片段在不同的哺乳动物序列中都存在保守的4个半胱氨酸位点用于二硫键的形成,并且都具有1个保守的糖基化位点。与α片段相似,在β片段中同样存在保守的3个半胱氨酸位点以及1个糖基化位点。并且人工饲养型诱导DQB基因序列发生了突变,在α片段中有5个位点发生突变,而在β片段中则有4个位点发生突变。

摘  要:
MHC-DQBα,βgene was cloned and sequenced and the differences with other mammals were found.By amino acid sequence alignment and phylogenetic analysis,we found that four Cys residues used to form a disulfide bond and one N-linked glycosylation site in different mammals inαdomain.Similarly,inβdomain we also found three Cys residues and one N-linked glycosylation site.In addition,the artificial breeding contributes to induce mutation of DQB gene.We found five mutations inαdomain and four mutations inβdomain.
