摘 要:
为测定不同叶龄枫香(Liquidambar formosana Hance)叶总黄酮的含量,取不同叶龄的枫香叶,用80%乙醇以料液比1∶10超声提取,以芦丁为参照物,采用三氯化铝比色法,在波长421 nm处用分光光度法测定其吸光度,依标准曲线计算幼嫩叶、成熟叶和落叶不同叶龄的叶片总黄酮含量。结果表明,幼嫩叶、成熟叶和落叶的总黄酮含量分别为2.66%、3.27%、0.31%。高效液相色谱测定结果显示,不同叶龄的枫香叶片黄酮类成分基本相同,但含量存在差异,其中落叶中黄酮含量最低。对嫩叶和成熟叶而言,成熟叶总峰面积要高于嫩叶峰面积约40%。因此,枫香叶中总黄酮含量与叶龄有关,成熟叶含量最高,嫩叶次之,落叶含量最低。在同等条件下,枫香叶处于成熟期时黄酮含量相对较高。在不考虑其他因素影响的情况下,成熟枫香叶是提取枫香天然黄酮的最佳资源。

The total flavonoids in leaves of Liquidambar formosana Hance at different stage were extracted by ultrasonic method using 80% ethanol with solid to liquid ratio at 1 ∶10,and then determined the absorbance at 421 nm by Al Cl3 colorimetry using rutin as control. The content of total flavonoids was calculated by the standard curve. The results showed that the content of total flavonoids in young leaves, mature leaves and defoliation was 2.66%, 3.27%, and 0.31%, respectively.According to characteristic peaks of high performance liquid chromatography, the composition of flavonoid compounds in L.formosana leaves at different stage was basically the same while the content was different. The content of flavonoids in defoliation was the lowest. As for young and mature leaves, the total peak area of mature leaf was about 40% higher than that of young leaves. Therefore, the content of total flavonoids in mature leaf is the highest, and followed by young leaves. In conclusion, the content of total flavonoids in mature leaves is the highest, thus are the best resource for extraction of natural food pigment.
