东亚飞蝗和刺腿食蚜蝇内生酵母菌多样性分析

摘  要:
为探究东亚飞蝗和刺腿食蚜蝇内生酵母菌的菌群多样性和探索这两种在农业生态系统中具有不同功能的昆虫内生酵母菌的多样性差异,通过Illumina MiSeq高通量测序技术对2个样本内生酵母菌26SrDNA进行测序,利用Mothur软件计算东亚飞蝗和食蚜蝇体内酵母菌的操作分类单元(operational taxonomic unit, OTU)并进行丰富度Alpha多样性分析,利用SPSS软件进行多样性差异显著性分析,共产生906个OTU,东亚飞蝗中含647个OTU,而食蚜蝇为677个OUT,共有OUT 418个;酵母菌共鉴定出2门19属。结果表明刺腿食蚜蝇内生酵母菌多样性略高于东亚飞蝗。东亚飞蝗体内发现了特有的掷孢酵母属(Sporobolomyces)和拟威尔嗜杀酵母(Cyberlindnera)生防酵母资源。新疆东亚飞蝗和刺腿食蚜蝇的内生酵母菌丰富多样,其生存的微环境具有特殊性。通过对其进一步研究可能为害虫防治和特殊环境下的酵母菌资源利用提供新思路。

摘  要:
To study the structural diversity of the endobiotic yeast isolated from Locusta migratoria manilensisand Ischiodon scutellaris Fabricius and compare diversity of endobiotic yeasts isolated from insdects with differenet ecologic functions inagriculture ecosystem, we sequenced 26 SrDNA of endobiotic yeast from two samples by Illumina Miseq high-throughput sequencing method, and classified operational taxonomic unit and analysed the Alpha diversity by Mouthor software. And then,we analysed the signifiance of diversity difference by SPSS, a total of 906 OTU were detected, 647 OTU from Locusta migratoria migratoria and 677 OTU from Ischiodon scutellaris Fabricius. among which 418 OTU are shared in two samples. Yeasts were identified and classified into 2 plylums and 19 genus in the two samples. The biodiversity of endobiotic yeas in Locusta migratoria migratoria is little more than that in Ischiodon scutellaris Fabricius. Meanwhile, we identified biological control yeasts belongs to Sporobolomyces and Cyberlindneraonly in Locusta migratoria migratoria The endobiotic yeasts isolated from Locusta migratoria migratoria and Ischiodon scutellaris Fabricius in Xinjiang are abundant and various. The micro-environment they existed is special. It could provide new ideas for pest control and yeasts resoueses utilization in special environment.
