高产谷胱甘肽酵母菌株的筛选和抗乙硫氨酸突变株的选育

摘  要:
[目的]选育高产谷胱甘肽的酵母菌株。[方法]通过紫外线进行诱变处理,运用推理育种技术获得抗乙硫氨酸突变株。[结果]从土壤中筛选出一株高产GSH的菌株HSJB1,其摇瓶发酵生物量为3.87 g/L,GSH产量为91.87 mg/L。依据菌株的细胞形态特征及生理生化特征,初步鉴定其为Saccharomyces cerevisiae。通过紫外线对出发菌株HSJB1进行诱变处理,获得一株抗乙硫氨酸突变株YBS77,该突变株经摇瓶发酵,生物量达7.60 g/L,GSH产量为211.96 mg/L。[结论]通过选育获得的突变株生物量比出发菌株提高了96.38%,GSH产量提高了130.72%,表明该育种方法可行。

摘  要:
[Objective]The purpose of the study was to breed high-production glutathione Saccharomyces.[Method] by UV and rational screening to obtain ethionine-resistant mutants.[Result] The strain HSJB1 which could accumulate high concentration of GSH was isolated from soil.The yield of GSH reached 91.87 mg/L and the yield of biomass reached 3.87 g/L.In addition to general physiological and biochemical properties,by the first step the results showed that the strain HSJB1 is Saccharomyces cerevisiae.A ethionine-resistant mutant YBS77 was obtained.The biomass yeild of the mutant reached 7.60 g/L and the glutathione productivity reached 211.96 mg/L by flask culture.[Conclusion] The biomass yeild of the mutant was 96.38%,higher than the original strain,and the glutathione productivity was 130.72% higher than the original strain.The breeding method was effective.
