适于宁夏枸杞RAPD分析的DNA高效提取方法

摘  要:
采用优化的CTAB法提取枸杞叶片基因组DNA。利用紫外分光光度计、琼脂糖凝胶电泳检测所得DNA的浓度和纯度,并进行了RAPD初步扩增分析。DNA浓度在300μg/mL,可达400μg/g(叶片鲜重)。OD260/OD280比值为1.44~1.6(理想值为1.8)。结果表明:优化CTAB法所提取枸杞总DNA的纯度高、完整、量大,能满足RAPD扩增,条带清晰、稳定,并且提取操作的程序快速、简便,试验成本低、效率高。优化的CTAB法高效提取的枸杞基因组DNA不需经氯化铯梯度离心或柱层析纯化,可以直接用于RAPD分析。

摘  要:
Optimize the use of the CTAB method to extract barbarum leaf genomic DNA.By UV spectrophotometer,agarose gel electrophoresis detection of DNA derived from the concentration and purity,and conducted a preliminary amplification RAPD analysis.DNA concentration 300 μg/mL,up to 400 μg/g(leaf fresh weight).OD260/OD280 ratio was 1.44~1.6(the ideal value was 1.8).The results showed that CTAB method by optimizing the total DNA extract of wolfberry high purity,integrity,and volume was very large,to meet the RAPD amplified bands were clear and stable,and extraction procedures for the operation of fast,easy,the experimental's cost was lower and efficiency was higher.Optimize the CTAB method and efficient extract Lycium barbarum genomic DNA without CsCl gradient centrifugation or column chromatography purification,can be directly used for RAPD analysis.
