圆环病毒2型抗体胶体金检测试纸条的初步研制及应用

摘 要：
为快速检测猪群中猪圆环病毒2型抗体,采用柠檬酸三钠还原法制备胶体金颗粒,并标记纯化的猪圆环病毒Cap蛋白和兔IgG,包被至玻璃纤维膜上制备金标垫,将纯化抗原和羊抗兔IgG分别包被至硝酸纤维素膜的检测线与质控线上,建立了检测猪圆环病毒2型抗体的胶体金免疫层析方法并组装检测卡。结果显示,组装的检测卡只与圆环病毒阳性血清发生特异性反应,阳性血清稀释10倍后仍然能够检出。用此检测卡与商品化ELISA试剂盒对100份临床血清进行平行检测,阳性率分别为89%和93%,两种方法符合率在95%以上。该方法操作简便、特异性强,可广泛应用于基层,也为圆环病毒疫苗效果评价提供了良好的试剂选择。

摘 要：
To detect specific antibodies of porcine circovirus type 2(PCV2)with gold immuno-chromatographic assay,purified PCV2 capid protein and rabbit IgG were labeled with colloidal gold prepared with the tri-sodium citrate reduction method,meanwhile capid protein and goat anti-rabbit IgG were striped in the test line position and quality control line position respectively. The results indicated that the assembled test card reacted exclusively with PCV2 positive sera and the test sensitivity was up-to 10 fold dilutions. The coincidence rate of this test card with commercial ELISA kid was up to 95%.The method was rapid,specific and simple in operation,and it could be extended at grass-roots.
