新农1号狗牙根愈伤组织胚性维持及增殖研究

摘  要:
以新农1号狗牙根胚性愈伤组织为材料,研究激素、外源添加物和蔗糖浓度对其愈伤组织胚性维持及增殖的影响。结果表明,2,4-D与6-BA的适宜浓度分别为2.0,1.0mg/L,浓度过低或过高均会导致胚性愈伤增殖率下降,且褐化率升高;在培养基中添加CuSO4可提高胚性愈伤组织的增殖率,但小植株形成强度降低;脯氨酸的适宜浓度为2.5mg/L,与不添加脯氨酸相比,增殖率提高了25.0%,小植株形成强度提高了22.97%;蔗糖浓度对胚性愈伤增殖影响不显著,当其为30g/L时,愈伤质量较好,且褐化率较低。综合分析可知,适宜胚性维持培养基为添加蔗糖30g/L、琼脂8g/L、2,4-D 2.0 mg/L、6-BA1.0 mg/L、CuSO42.5 mg/L、脯氨酸2.5 mg/L、谷胱甘肽2.0mg/L的MS培养基;在此培养基中,胚性愈伤组织增殖率可达到75.0%,小植株形成强度为31.3%,褐化率较低,愈伤组织呈致密的淡黄色颗粒,胚性维持状况良好。

摘  要:
With embryogenic callus as materials,the effects of hormone,additives and sucrose on embryogenic maintenance and proliferation of Cynodon dactylon(L.)Per cv.Xinnong No.1were studied.The results showed that the suitable 2,4-D and 6-BA concentrations were 2.0mg/L and 1.0mg/L higher or lower concentration would lead to embryogenic callus proliferation rate decreasing and browning rate increasing.The culture medium added with CuSO4 could further improve the embryogenic callus proliferation rate,but reduce plantlets formation strength.The suitable concentration of proline was 2.5 mg/L,compared with proline,the proliferation rate increased by 25.0%,plantlets formation strength increased by 22.97%.The effect of sucrose on embryogenic callus proliferation was not obvious,when the concentration of sucrose was 30g/L,callus quality was better and favorable for plantlet regeneration.Comprehensive analysis showed that MS medium added with 30g/L sucrose,8g/L Agar,2.0 mg/L 2,4-D,1.0 mg/L 6-BA,2.5mg/L CuSO4,2.5mg/L Proline and 2.0mg/L GSH was most suitable,the embryogenic callus proliferation rate could reach 75.0%,Plantlet formation intensity was 31.3% and browning rate was low,Embryogenic callus was maintained in good condition and showed dense yellow particles.
