美系獭兔和伊拉肉兔FABP4基因多态性及其与肌肉脂肪酸含量的相关性

摘  要:
【目的】探寻影响兔肉中脂肪酸性状的优势基因型。【方法】利用PCR-RFLP技术检测60只美系獭兔、51只伊拉肉兔FABP4基因的单核苷酸多态性,分析其与兔肉中脂肪酸含量的相关性。【结果】2个兔品种在intron1中存在C/T突变,该突变位点能被TaiⅠ限制性酶识别,且经酶切后发现,2个兔群体中存在CC、CT和TT 3种基因型。美系獭兔的3种基因型频率分别为0.083,0.467和0.450,等位基因C和T的基因频率分别为0.683和0.317;伊拉肉兔的3种基因型频率分别为0.137,0.549和0.314,等位基因C和T的基因频率分别为0.588和0.412。关联分析结果表明,在美系獭兔群体中,CC型个体肌肉组织中的脂肪酸C14:0含量和脂肪酸C18:1,n-9含量显著高于CT和TT型个体;TT型个体脂肪酸C18:3,n-3含量显著高于CC和CT型个体;TT型个体脂肪酸C20:4,n-6含量显著高于CC型个体。在伊拉肉兔群体中,CC基因型个体肌肉组织中的C18:1,n-9含量显著高于CT和TT基因型个体(P<0.05);CC型个体脂肪酸C18:2,n-6含量显著高于CT型个体;TT型个体脂肪酸C18:3,n-3含量显著高于CC和CT型个体;CT型个体脂肪酸C20:4,n-6含量显著高于CC型个体。【结论】FABP4基因可作为美系獭兔和伊拉肉兔脂肪酸性状选育的候选基因。

【Objective】This study aimed to find the dominant genotype influencing fatty acid character in rabbit meat.【Method】FABP4gene single nucleotide polymorphism of 60Rex rabbits and 51IRA rabbits was detected with PCR-RFLP,and the correlation between SNP and fatty acid content of the two rabbits was analyzed.【Result】The intron1C/T mutation existed in both rabbit breeds,which can be recognized by TaiⅠrestriction enzymes.There were three genotypes:CC,CT,and TT.The genotype frequencies of Rex were 0.083,0.467and 0.450,and the allele gene frequencies of C and T were 0.683and 0.317.The genotype frequencies of IRA were 0.137,0.549and 0.314,and the allele gene frequencies of C and T were 0.588and 0.412.Correlation analysis showed that for Rex rabbits,the content of fatty acid C14:0and C18:1,n-9in CC genotype was significantly higher than that of CT and TT genotypes(P<0.05);C18:3,n-3contents of CC and CT were significantly lower than that of TT genotype;and C20:4,n-6content of TT genotype was significantly higher than that of CC.For IRA rabbits group,the fatty acid C18:1,n-9content of CC genotype was significant higher than that of CC and CT genotypes(P<0.05);C18:2,n-6content of CC genotype was significantly higher than that of CT genotype;C18:3,n-3content of TT genotype was significantly higher than that of CT and CC genotypes;and C20:4,n-6content of CT genotype was significantly higher than that of CC genotype.【Conclusion】FABP4gene can be used as candidate gene for fatty acids character of Rex and IRA rabbits.
