大果榉组织培养研究

摘  要:
试验以大果榉茎段为外植体,研究其最佳培养基配方及培养条件,为建立大果榉快繁体系提供理论依据。结果表明,茎段诱导不定芽在WPM+1.0 mg/L 6-BA+0.1~0.5 mg/L NAA培养基上生长良好,腋芽萌发率较高,有效芽较多,且生长健壮;大果榉茎段增殖的最佳培养基为WPM+1.0 mg/L 6-BA+0.5 mg/L NAA;生根培养最佳培养基为WPM+0.02 mg/L 6-BA+0.05 mg/L IBA,试管苗生根早,且根系发达。

摘  要:
Zelkova sinica is endemic to China with high afforestation value and ornamental value. The stem segments of Zelkova sinica were used as explants to study the optimum medium formulation and culture conditions, so as to provide theoretical basis for the establishment of rapid propagation system of Zelkova sinica. The results showed that the adventitious buds which was induced from stem segments grew well under the treatment of WPM+1.0 mg/L 6-BA+0.1-0.5 mg/L NAA, with high budding rates, more effective buds and grow strong. The best cultural medium for propagation of Zelkova sinica by using stem segments was WPM+1.0 mg/L 6-BA+0.5 mg/L NAA. The optimum medium for rooting culture was WPM+0.02 mg/L 6-BA+0.05 mg/L IBA, and the rooting of test-tube seedlings was early and the roots were developed.
