长春花法尼基转移酶β-亚基基因的克隆及其在黄龙病菌感染过程中的表达分析

摘  要:
为了探讨长春花法尼基转移酶β-亚基(crFTB)在黄龙病菌侵染过程中的表达模式及作用,采用RACE技术扩增长春花crFTB基因的cDNA全长序列,并进行生物信息学分析,采用实时荧光定量PCR技术检测叶片组织中crFTB在黄龙病菌(CLas)侵染长春花不同时间点的表达变化.结果显示,crFTB cDNA全长为1826 bp,包含150 bp 5′UTR、308 bp 3′UTR和1368 bp开放阅读框,编码455个氨基酸,理论分子质量和等电点分别为50.45 ku和4.67.FTB蛋白在物种间有较好的保守性,系统进化树表明,crFTB与油橄榄樟子松变种FTB蛋白的亲缘关系最近.CLas侵染长春花过程中,crFTB在叶片组织中的表达量呈现先升后降的模式,从接种后4 d开始crFTB基因的表达量升高,到24 d时达到峰值,然后表达量开始下降,直到试验结束时仍显著高于健康植株中crFTB基因的表达量.分析认为,crFTB在长春花抵抗CLas侵染过程中起重要作用.

摘  要:
To study the expression pattern and role of farnesyltransferase β-subunit in Catharanthus roseus(crFTB) during the infection of Candidatus Liberibacter asiaticus(CLas),the full-length cDNA sequence of crFTB was obtained by RACE,and expression pattern of crFTB in C.roseus during CLas-infection was analyzed at different time by quantitative real-time PCR technology.The full-length cDNA of crFTB was 1 826 bp,containing 150 bp 5′UTR,308 bp 3′UTR,and 1 368 bp ORF,which encoded 455 amino acids.The molecular weight and isoelectric point were predicted to be 50.45 ku and 4.67 respectively.Multi-sequence comparison showed that FTB among different species was highly conserved,and phylogenetic relationship analysis revealed that crFTB protein had close genetic relationship with FTB of Olea europaea var.sylvestris.During CLas-infection,crFTB expression level in leaves showed a pattern of rising first and then falling.The crFTB expression level increased gradually and reached the highest value during 4 to 24 days after inoculation,and then decreased gradually,but still higher than the expression level of healthy plants till the end of the experiment.By analysis,the crFTB plays an important role during the resistance of C.roseus against CLas.
