油菜籽硫葡萄糖甙速测技术的研究——3,5-二硝基水杨酸分光光度法

摘  要:
培育油菜低芥酸及低硫葡萄糖甙的“双低”品种是当前我国油菜工作者的一个重要课题。我省也正在进行着这个方面的研究工作。但在着手这项工作中所遇到的第一个问题便是,检测起来很困难,至今还没有能够找到有关它们的速测技术,这就给工作的顺利开展造成了难以克服的障碍。为解决这个问题,最近,芥酸的速测技术已为福建省农科院解决,实现了“油菜籽切片法及芥酸气液色谱法”,宣告芥酸速测技术已在我国研究成功。本文则提出“油菜籽硫葡萄糖甙3,5-二硝基水杨酸法”,是我农学院及省农科院的共同研究成果,同时宣告硫葡萄糖甙速测技术也进一步在我国研究成功。

摘  要:
Glucosinolates in rapeseed can be rapidly determined by using 3,5-dinitrosalicylic acid spactrophotometric method. The sample to be analyzed is weighed out two equal portions.One portion of the sample after being boiled with water is used for glucose determination, and the another portion after being completely hydrolyzed in 37℃ water bath and then boiled with water is used for glucose and glucosinolates-glucose determination. The loss in glucose from glucose and glucosinolates-glucose represents glucosinolates-glucose.Coefficients for converting glucosinolates-glucose into iso-thiocyanate (or thiocyanate) and glucosinolates are taken to be 0.5499 and 2.207 respectively.
