高效液相色谱法测定鳗鲡各组织中的乙酰甲喹残留量

摘  要：
建立鳗鲡各组织中乙酰甲喹残留量的高效液相色谱测定方法,高效液相色谱检测条件为:采用SunFireTM C_(18)柱(4.6mm×150mm,3.5μm),柱温35℃;以甲醇∶水(40∶60,体积比)为流动相,流速为0.8mL·min~(-1);采用二极管阵列检测器,检测波长239nm。结果表明:在0.05~2.5μg·mL~(-1)范围内,乙酰甲喹具有良好的线性关系,含量与峰面积的相关系数达0.999以上;乙酰甲喹在鳗鲡肌肉、肾、肝和血浆中的检出限分别为3μg·kg~(-1)、15μg·kg~(-1)、15μg·kg~(-1)、15μg·L~(-1),鳗鲡各组织中乙酰甲喹回收率在76%~90%,相对标准偏差均小于10%(n=6)。本方法简便、快捷、灵敏度高,具有良好的重复性,满足鳗鲡各组织基质内乙酰甲喹残留量的快速定量分析的要求。

摘  要：
An HPLC methodology was developed for determining mequindox residues in the organs of eels.The analysis was conducted under the following conditions:SunFireTM C_(18)column(4.6 mm×150 mm,3.5μm)at35℃,methanol:deionized water(v/v 40∶60)for themobile phase with a flow rate of 0.8mL·min~(-1),and 239 nm wavelength seton PDA detector for optical density measurements.With in the mequindox concentration range of0.05-2.5μg·mL~(-1),an excellent linear regression between the drug concentration and the peak area under curve was obtained with a correlation coefficient greater than 0.999.LODs of mequindox in the muscle,liver,kidney,and plasma from the eelswere 3μg·kg~(-1),15μg·kg~(-1),15μg·kg~(-1),and 15μg·L~(-1),respectively.The average mequindox recovery rate of the method ranged from 76% to 90% with RSD(n=6)below 10%.It was,thus,concluded that the newly developed simple,rapidmethodology was highly sensitive and repeatable in detecting mequindox residuein the tissues of the eels.
