不同倍性葡萄种质激素水平比较分析

摘  要:
[目的]通过分析3组同基因型不同倍型鲜食葡萄种质植株中3种内源激素含量,为葡萄倍性育种以及栽培生理的研究提供借鉴。[方法]以3份四倍体鲜食葡萄种质(‘四倍体红地球’、‘四倍体玫瑰香’和‘2-2-4’)及其二倍体种质‘红地球’、‘玫瑰香’和‘2-2-2’一年生营养袋扦插苗新梢和根系为材料,利用HPLC多阶质谱检测对新梢和根系中的生长素(IAA)、脱落酸(trans-ABA和cis-ABA)和10种细胞分裂素含量进行测定,后续定量分析选择离子检测方式选定各激素裂解后的特征碎片离子。[结果]3组材料中四倍体葡萄叶片中IAA、ABA含量均显著低于相应的二倍体,IAA含量为二倍体葡萄的38. 2%~77. 9%,ABA含量为二倍体葡萄的23. 6%~80. 0%。‘四倍体红地球’叶片中有7种细胞分裂素含量显著高于二倍体‘红地球’,为后者的1. 36~3. 99倍;‘四倍体玫瑰香’叶片中仅DHZR显著高于二倍体‘玫瑰香’,为后者的1. 49倍;而‘2-2-4’中9种细胞分裂素含量均显著低于‘2-2-2’,为后者的12. 7%~68. 6%。3组材料四倍体葡萄根系中仅‘四倍体红地球’IAA和c-ABA含量显著高于二倍体‘红地球’,‘四倍体玫瑰香’ABA含量显著低于二倍体‘玫瑰香’。根系中,‘四倍体红地球’中有5种细胞分裂素显著低于‘红地球’。‘2-2-4’ZOG和DHZR含量显著高于‘2-2-2’,为后者的1. 26、3. 82倍,ZR、iP和iPR含量则显著低于‘2-2-2’。‘四倍体玫瑰香’中ZOG含量显著高于‘玫瑰香’,而ZR、DHZR、iP和iPR4种细胞分裂素则显著低于‘玫瑰香’。[结论]二倍体葡萄加倍后,植物体内源激素的含量未因染色体加倍而呈现规律性加倍。

摘  要:
[Objective]This study was designed to analyze the content of three endogenous hormones in three groups of table grape of isogenic genotypes and different ploidy germplasm,with the view to providing scientific reference for the study of grape ploidy breeding and cultivation physiology. [Methods]The new shoots and roots of cutting seedlings of three copies of tetraploid table grape germplasm including "hongsiqiu", "meihuixiang" and "2-2-4",and their diploid germplasm named as "hongdiqiu", "meiguixiang" and "2-2-2" were used as experimental materials. The contents of auxin(IAA),abscisic acid(trans-ABA and cis-ABA)and 10 different cytokinins were measured by HPLC multi-step mass spectrometry,and the subsequent quantitative analysis selected ion detection method was used to select the characteristic fragment ion after the cleavage of each hormone.[Results]The contents of IAA and ABA in the leaves of the tetraploid grape of these three groups were significantly lower than that of the corresponding diploids. The content of IAA from tetraploid was determined as 38. 2% ~ 77. 9% of the diploid,and the content of ABA from tetraploid was 23. 6% ~ 80. 0% of the diploid grapes. The content of seven cytokinins in the leaves of tetraploid "hongdiqiu" were significantly higher than that of its diploid,which was around 1. 36~3. 99 times of the latter.And only DHZR in the leaves of tetraploid "meiguixiang" was significantly higher than its diploid,which was around 1. 49 times that of the latter. The content of nine Cytokinins in the leaves of "2-2-4" was statistically lower than "2-2-2",which was around12. 7%~68. 6% of the latter. In the roots of three groups of tetraploid grapes,only the contents of IAA and c-ABA in the roots of tetraploid "hongdiqiu" were significantly higher than that in diploid individual,and ABA content in the roots of tetraploid "meiguixiang" was significantly lower than that in diploid counterpart. As for the content of Cytokinins in the roots,five kinds of Cytokinins in the tetraploid "hongdiqiu" were statistically lower than those in the diploids. The content of ZOG and DHZR in 2-2-4 was significantly higher than that in 2-2-2,which were 1. 26 and 3. 82 times of the latter,and the content of ZR,iP and iPR was significantly lower than that of 2-2-2. and the content of ZOG in tetraploid "meiguixiang" was higher than that of diploids,while the contents of ZR,DHZR,iP and iPR4 were significantly lower than diploid individuals. [Conclusion]When the chromosomes of diploid grape was doubled,the content of the endogenous hormones in the plant did not double regularly. Due to the complexity of the polyploid,the relationship between the increase of chromosome multiples and the change of the amount of the endogenous hormone is very complicated,and it is of great significance to study for better understanding the mechanisms from different sides.
