H9亚型禽流感病毒免疫磁珠分离方法初步建立

摘  要:
禽流感病毒(AIV)严重危害世界养禽业,是举世瞩目的重大人畜共患病原,环境中微量病原是潜在的重要传染源,很难进行监测。为了提高畜禽环境中微量病原的监测效率,样品的必要预处理非常重要。制备了粒径约30~100 nm的Fe_3O_4纳米粒子,并利用戊二醛二步法偶联H9-HA单抗和磁珠,偶联效率约为130 mg/g;200μL免疫磁珠(5.6 mg/m L)可分离纯化100μL血凝价为28的AIV-H9。制备的免疫磁珠可以为纯化和富集畜禽生活环境中的微量病原提供便利,结合其他病原检测手段将有助于提高环境中病原的监测效率。

摘  要:
Avian influenza virus(AIV) was one of the major zoonotic pathogens which seriously harmful to the world poultry industry. AIV is excreted in the environment by AIV-infected birds and the trace pathogens were the potentially important source of infection but difficult to be monitored. In order to effectively monitoring the pathogens in poultry environment, the pretreatment for samples was very important. In this study, Fe_3O_4 nanoparticles with particle size about 30-100 nm were prepared, and the coupled with H9-HA monoclonal antibody and beads. The coupling efficiency was about 130 mg / g. The results showed that 200 μL immunomagnetic beads(IMB) at concentration about 5.6 mg / m L could isolate and purify 100 μL of AIV-H9 with hemagglutination titer for 28. It indicated that IMB could provide convenience to purify and enrich trace pathogens in of animal living environment, which will improve the efficiency of monitoring pathogens in the environment cooperating with other method.
