超临界CO_2萃取制备远志籽油工艺的优化及其成分鉴定

摘 要:
利用远志籽为原料,采用超临界CO_2萃取技术,选择对远志籽油提取率影响较大的三种因素:萃取压力、萃取时间及乙醇夹带剂添加量,以提取率为考察指标进行正交试验,优化远志籽油的提取工艺;按照国标方法对制备的远志籽油的理化特性进行测定分析;采用薄层色谱法对远志籽油的甘油三酯结构进行初步鉴定;通过气相色谱法对远志籽油的脂肪酸成分进行分析。结果表明,远志籽的含油量为35. 24%;超临界CO_2萃取法萃取远志籽油的最佳条件为萃取压力30 MPa,萃取时间5 h,乙醇夹带剂用量25%,在此条件下远志籽油的提取率为95. 6%;制备的远志籽油的酸价为1. 79 mg KOH/g,过氧化值为5. 75 mmol/kg,其基本理化特性符合国家四级食用油的标准。通过薄层色谱法初步鉴定,远志籽油中含有甘油三酯和sn-2-乙酰基甘油三酯两种组分;气相色谱法对远志籽油的脂肪酸成分分析结果表明,其籽油含有6种脂肪酸成分,其中油酸和花生一烯酸含量较高,分别为31. 77%和49. 46%,不饱和脂肪酸含量为82%。该研究结果将为远志籽油的开发利用提供参考依据。

Using polygala seeds as raw materials,the supercritical CO_2 extraction technology was optimized by selecting three factors influencing extraction rate of polygala seed oil: extraction pressure,extraction time and ethanol entrainer addition amount. The orthogonal test was carried out with the extraction rate as an indicator to optimize the three extraction factors. The physical and chemical properties of polygala seed oil were determined according to the national standard methods. The structure of triglyceride in polygala seed oil was preliminarily identified by thin layer chromatography. The fatty acid composition of polygala seed oil was determined by gas chromatography. The results showed that the oil content of polygala seed oil was 35. 24%.The orthogonal test results showed that the optimum conditions for extracting polygala seed oil were extraction pressure as 30 MPa,extraction time as 5 hours,and ethanol entrainer dosage as 25%. Under these conditions,the extraction rate of polygala seed oil was 95. 6%. The acid value of polygala seed oil was 1. 79 mg KOH/g,and the peroxide value was 5. 75 mmol/kg. Its basic physical and chemical properties were in line with the national fourth-grade edible oil standard. The TLC analysis determined the presence of triglyceride and sn-2-acetyl triglyceride in polygala seed oil. By analyzing the fatty acids of polygala seed oil by gas chromatography,a total of six fatty acid components were obtained,among which,the content of oleic acid and peanut monoenoic acid were higher,and the contents were 31. 77% and 49. 46%,respectively; the unsaturated fatty acid content was 82%. The results provided a reference for the development and utilization of polygala seed oil.
