静原鸡FOXL2基因PCR扩增及生物信息学分析

摘  要:
为研究FOXL2基因对鸡的性成熟及产蛋性能的影响,试验以优良地方品种静原鸡为研究对象,利用PCR技术成功克隆了静原鸡心脏FOXL2基因的CDS区,其序列长度为918 bp。生物信息学方法分析结果显示,静原鸡FOXL2基因编码了305个氨基酸,分子式为C_(1491)H_(2267)N_(425)O_(432)S_(15)。FOXL2蛋白是亲水性蛋白质,但不存在跨膜区域,不含信号肽。FOXL2蛋白含有28个丝氨酸(Ser)磷酸化位点,含有4个苏氨酸(Thr)磷酸化位点,含有16个酪氨酸(Tyr)磷酸化位点。FOXL2蛋白在第46~132位氨基酸之间只有1个Forkhead结构。FOXL2蛋白二级结构主要的结构形式为α-螺旋与无规则卷曲。在SWISS-MODEL中建立FOXL2蛋白的三级结构,模型以2ef0.1.A(Ornithine Carbamoyltransferase)蛋白为模板,模板的序列同源性高达99.34%。与其他禽类FOXL2基因CDS区序列进行多序列比对,使用MEGA 7.0软件构建进化树。静原鸡与环颈雉亲缘关系较近,与原鸡的亲缘关系最近。

摘  要:
In order to study the effect of FOXL2 gene on the sexual maturity and egg production performance of chicken,the study took the excellent local breeds tof jungle-bush as the research object,and successfully cloned the CDS region of FOXL2 gene of jungle-bush heart by using PCR technology,with the sequence length of 918 bp.The results of bioinformatics analysis showed that the FOXL2 gene encoded 305 amino acids,with the molecular formula C_(1491)H_(2267)N_(425)O_(432)S_(15).FOXL2 protein is a hydrophilic protein,but there is no transmembrane region and no signal peptide.The FOXL2 protein contains 28 serine(Ser)phosphorylation sites,4 threonine(Thr)phosphorylation sites,and 16tyrosine(Tyr)phosphorylation sites.The FOXL2 protein has only one Forkhead structure between amino acids 46~132.The primary secondary structure of FOXL2 is helical and irregularly coiled.The three-level structure of FOXL2 protein was established in swiss-model,with 2ef0.1.a(Ornithine carbamoyltransferase)protein as the template,and the sequence homology of the template was as high as 99.34%.Ncbi-blast was compared with the CDS region sequences of FOXL2gene in other birds,and the evolutionary tree was constructed by using the software MEGA 7.0.It can be seen that the prairie chicken is closely related to the ring-napped pheasant and closest to the jungle fow.
