三丁基锡(TBT)对罗非鱼精巢细胞凋亡的影响

摘  要:
以雄性奥利亚罗非鱼为试验材料,研究了三丁基锡(TBT)对鱼类精巢细胞凋亡的影响。通过腹腔注射染毒TBT(剂量分别为0、1、3、5、10μg·kg-1体重),取染毒后的精巢,一部分用于制备石蜡切片,原位末端标记法(TUNTEL法)进行细胞凋亡分析,另一部分用于研究TBT对精巢细胞Ca2+-ATP酶活性的影响。结果表明,染毒后24和48h,随着染毒剂量的升高,罗非鱼精巢细胞凋亡率先增高后降低,在5μg·kg-1体重时达最高;染毒后96h,相应细胞凋亡率先降低后升高,在5μg·kg-1体重时最低。与之相对应的精巢细胞Ca2+-ATP酶活性,低剂量组(1~3μg·kg-1)与对照组相比,差异不显著;高剂量组与对照组相比,差异显著或极显著;在试验剂量范围内,Ca2+-ATP酶活性随着注射剂量的升高而升高。

摘  要:
The effect of tributyltin chloride (TBT) on apoptosis of fish spermary cell was researched by using male Oreochromis aureus as experiment material. TBT was injected intraperitoneally into Oreochromis aureus (dosages were 0,1,3,5,10 μg·kg-1 body weight,respectively),and the spermary of fish injected was sampled. One part of spermary was used to make paraffin slice,using terminal deoxynucleotidy transferase mediated dUTP-Biotin nick-end labeling (TUNTEL) method to analyze apoptosis. The other was used to study the influence of TBT on Ca2+-ATP enzyme in the spermary cell. The result showed that with the increase of injection dosage,the spermary cell apoptosis rate of tilapia increased firstly and then reduced after 24 and 48 hours of injection and the apoptosis rate was highest when the dosage was 5 μg·kg-1 body weight. However,the cell apoptosis rate reduced firstly and then increased after 96 hours of injection and the apoptosis rate was lowest when the dosage was 5 μg·kg-1 body weight. No remarkable difference was observed in the activities of Ca2+-ATP enzyme in spermary cell between low dose group (1~3 μg·kg-1) and the control group,but the difference between the high dose group and the control group was remarkable or extremely remarkable. The activity of Ca2+-ATP enzyme increased with the experimental dosage of 0~10 μg·kg-1.
