高效液相色谱法测定中药复方透皮软膏中绿原酸和咖啡酸的含量

摘 要:
建立了同时测定中药复方透皮软膏中绿原酸和咖啡酸含量的高效液相色谱方法。采用Shim-pack VP-ODS(150 mm×4.6 mm,5μm)色谱柱,以乙腈和0.1%磷酸溶液(体积比为9∶91)为流动相,等度洗脱,327 nm处检测,绿原酸和咖啡酸分别在0.078~0.786μg/mL(R2=0.999 7)和0.042~0.420μg/mL(R2=0.999 5)范围内线性关系良好,平均回收率分别为98.95%和100.06%,RSD分别为0.42%和1.35%。结果表明,此方法灵敏度高、操作简便、结果准确、重复性好,可用于中药复方透皮软膏中绿原酸及咖啡酸含量测定。

摘 要:
To determinate the contents of chlorogenic acid and caffeic acid in Chinese herbal medicine compound percutaneous ointment,a high performance liquid chromatograph(HPLC) method was established.Two compounds were analyzed using a HPLC system equipped with Shim-pack VP-ODS(150 mm × 4.6 mm,5 μm) column.Acetonitrile-0.1% phosphoric acid solution(9/91,V/V) was used as the mobile phase.The isolated gradients were detected at 327 nm wavelength with an ultraviolet detection.The results showed that the method had good linear relationship in the range of 0.078~0.786 μg/mL(R2=0.999 7) for chlorogenic acid and 0.042~0.420 μg/mL(R2=0.999 5) for caffeic acid,respectively.The average recovery was 98.95% with RSD of 0.42% for chlorogenic acid and 100.06% with RSD of 1.35% for caffeic acid.The study showed that this method is sensitive,convenient,accurate and reproducible for determining the contents of chlorogenic acid and caffeic acid in Chinese herbal medicine compound percutaneous ointment.
