苦豆子内生放线菌产酶特性分析及活性菌株鉴定

摘  要:
以宁夏干旱荒漠区的126株苦豆子为试材,采用透明圈法对分离自其内生放线菌进行分泌胞外酶活性检测。结果表明:有71株内生放线菌具有酶活性,占总菌株数的56.3%。其中有3株菌株酶活性最为显著,菌株NDZKDS73、NDZKDS7具有的淀粉酶活性最高,透明圈半径达到24.62mm和19.12mm;菌株FJYKDS5具有的蛋白酶活性高,透明圈半径16.94mm;菌株FJYKDS11、BJGKDS2具有的纤维素酶活性最高,透明圈半径为24.72mm和22.40mm;菌株FJJKDS2具有的几丁质酶活性最高,透明圈半径17.76mm;菌株NDZKDS24、NDZKDS20对葡聚糖酶活性最高,透明圈半径为12.06mm和10.50mm。通过形态特征和16S rRNA基因序列同源性分析,有7株活性菌属链霉菌属(Streptomyces),1株属拟诺卡氏菌属(Nocardiopsis)。

摘  要:
Taking 126 endophytic actinomycetes from Sophora alopecuroides L.in arid desert region of Ningxia as material,through transparent circle method,studied the feature of excreting enzyme.The results showed that the 71 strains could produce enzyme,accounting for 56.3%of total isolates,in which 8 strains were the most significantly active.In these strains,NDZKDS73 and NDZKDS7 strains produced the highest activity amylase,the transparent circle radius were 24.62 mm and 19.12mm,respectively;FJYKDS5 strain produced the highest activity protease,the transparent circle radius was 16.94 mm;FJYKDS11and BJGKDS2 strains produced the highest activity cellulase,transparent circle radius were 24.72 mm and 22.40 mm,respectively;FJJKDS2 strains produced the highest activity chitinase,transparent circle radius was 17.76 mm;NDZKDS24 and NDZKDS20 strains produced highest activity glucanase,transparent circle radius were 12.06 mm and 10.50 mm,respectively.By morphological characteristics and 16S rRNA gene sequence similarity analysis,7 strains were identified as Streptomyces and 1 strain were Nocardiopsis in 8 strains.
